Modulation of immune responses by DNA damage signaling

چکیده

An accumulation of evidence indicates the importance DNA damage signaling in modulating immune responses. Indeed, understanding mechanism that underlies signal transduction originating from is vital to overcoming refractory cancer, particularly when cancer therapy applied combination with damage-dependent radio/chemotherapy. In addition, immune-associated responses such signals can aggravate symptoms infections, allergies, autoimmune disease, and aging. this review, we discuss how cells transduce signals, triggered by damage, their origins neighboring affects inflammatory

Modulation of macrophage immune responses by Echinacea.

Echinacea preparations are widely used herbal medicines for the prevention and treatment of colds and minor infections. There is little evidence for the individual components in Echinacea that contribute to immune regulatory activity. Activity of an ethanolic Echinacea extract and several constituents, including cichoric acid, have been examined using three in vitro measures of macrophage immun...

Taming the tiger by the tail: modulation of DNA damage responses by telomeres

Telomeres are by definition stable and inert chromosome ends, whereas internal chromosome breaks are potent stimulators of the DNA damage response (DDR). Telomeres do not, as might be expected, exclude DDR proteins from chromosome ends but instead engage with many DDR proteins. However, the most powerful DDRs, those that might induce chromosome fusion or cell-cycle arrest, are inhibited at telo...

Activation of STING-Dependent Innate Immune Signaling By S-Phase-Specific DNA Damage in Breast Cancer

Background Previously we identified a DNA damage response-deficient (DDRD) molecular subtype within breast cancer. A 44-gene assay identifying this subtype was validated as predicting benefit from DNA-damaging chemotherapy. This subtype was defined by interferon signaling. In this study, we address the mechanism of this immune response and its possible clinical significance. Methods We used i...

Comparative Evaluation of Mx and Alum as Bio and Conventional Adjuvants in Inducing Immune Responses by Influenza DNA Vaccine

Background & Objective: DNA vaccines as a new generation of vaccines require adjuvant to improve vaccine immunogenicity; adjuvants can also increase the DNA vaccine efficacy. In this study, the effects of the host’s interferon-inducible Mx protein as bio adjuvant and conventional alum adjuvant were evaluated.   Materials & Methods: The BALB/c mice were immunized by different prime-boost strate...

DNA damage response inhibition at dysfunctional telomeres by modulation of telomeric DNA damage response RNAs

The DNA damage response (DDR) is a set of cellular events that follows the generation of DNA damage. Recently, site-specific small non-coding RNAs, also termed DNA damage response RNAs (DDRNAs), have been shown to play a role in DDR signalling and DNA repair. Dysfunctional telomeres activate DDR in ageing, cancer and an increasing number of identified pathological conditions. Here we show that,...
